Abstract:

 【Abstract】 Objective To investigate changes of the expression of rat cortical GLUT3 mRNA and protein during cardiopulmonary resuscitation of cardiac arrest. Methods Thirty of SD rats were randomly divided into sham operation group(S group,n=5), resuscitation group (R group, n=25),and resuscitation group was further divided into five subgroups at restoration of spontaneous circulation(ROSC) 3,6,12,24,48 h. The rat model of asphyxial cardiac arrest and cardiopulmonary resuscitation (CPR) was established and the animals were sacrificed at each time point after CPR,and brain tissues were taken..Real-time qPCR and immunohistochemistry staining were performed to evaluate cortical GLUT3 expression of parietal lobe. Results In R group, GLUT3 increased gradually at ROSC 3 h,peaked at ROSC6 and 24 h, while dropped at ROSC 12 h. An upregulation of GLUT3 still existed at ROSC 24 h,although dropped at ROSC 12 h. Conclusion GLUT3 expression increased after cerebral ischemia and hypoxia caused by cardiac arrest and CPR, which may be a protective response to ischemic injury.

【摘要】 目的 研究在心搏骤停心肺复苏过程中大鼠脑皮质葡萄糖转运体3(GLUT3)转录水平和蛋白表达水平的变化。方法 30只大鼠随机分为两组,假手术组(S,n=5),心肺复苏组(R,n=25),心肺复苏组分别选取自主循环恢复(ROSC)3、6、12、24、48 h 5个时间点为5个小组,用窒息法制备稳定的大鼠心搏骤停及心肺复苏模型。R组在ROSC 3、6、12、24、48 h取大脑双侧顶叶皮质,分别用实时荧光定量PCR及免疫组织化学半定量方法(SP法)检测相应时间点GLUT3mRNA和蛋白的表达。结果〓R组与S组相比较,GLUT3mRNA水平在ROSC3h出现轻微升高,在ROSC 6 h及24 h达高峰,12 h GLUT3mRNA表达水平出现一下降点,但仍高于S组,ROSC48h GLUT3mRNA仍高于假手术组。GLUT3蛋白表达的变化趋势与GLUT3mRNA的变化一致。结论 在心搏骤停心肺复苏过程中,所造成的全脑缺氧缺血状态会使大鼠脑皮质GLUT3的表达水平上调,可能是机体对缺血损伤的保护性反应。
