The application of the primary culture of liver cancer cells with tissue slice method

Abstract: 【Abstract】〓Objective〓To investigate the application of the primary culture of liver cancer cells with tissue slice method. Methods〓The primary liver cancer cells from human liver tissue were isolated by using tissue slice method and tissue pieces respectively, and the success rate of cultivation, the cell climbed-out time, cell morphology, and the expression of GPC3 of two methods were analyzed. Results The success rate of cultivation with the tissue slice method was 66.67%,the cell climbed-out time was (7.89±0.78)..d..In the second generation of culture cells can be observed with morphological rules. Meanwhile HepG2 cells and primary liver cancer stably expressed GPC3..Conclusion The both methods can successfully cultured primary liver cancer cells,.but it is better with tissue slice method to raise the liver cancer cells in the cell morphology and cleanliness and which can stably express GPC3

摘要: 【摘要】〓目的〓探讨组织薄片法在人原代肝癌细胞培养中的应用。方法〓分别运用组织薄片法和组织块法从人肝癌组织标本中分离培养原代肝癌细胞,分析两种方法培养成功率、细胞爬出时间、形态及GPC3的表达。结果〓薄片法培养成功率为66.67%,爬出细胞时间为(7.89±0.78)d,在培养第一代就能观察到形态规则的细胞且周围培养环境洁净;组织块法培养成功率为61.11%,爬出细胞时间为(7.56±0.73)d,在培养第二代能观察到形态规则的细胞。同时原代肝癌细胞和HepG2均能稳定表达GPC3。结论〓两种方法均能成功培养出原代肝癌细胞,但薄片法培养的细胞在形态及洁净程度上较组织块好,同时能稳定表达GPC3。
