Expression and clinical significance of Polo?like kinase 3 in prostate cancer

摘要: 目的 探讨Polo样激酶3(Polo?likekinase3,Plk3)在前列腺癌组织中的表达情况及其临床意义。方法 采用免疫组化技术检测68例前列腺癌组织及10例正常前列腺组织中Plk3的表达情况,分析Plk3表达与临床病理特征之间的关系,并通过对TCGA数据库中498例前列腺癌患者的Plk3表达值进行GSEA分析。结果 Plk3在前列腺癌组织中比癌旁组织中表达阳性率高,且差异有统计学意义(P=0.034);Plk3表达与前列腺癌的病理分级、临床分期、T分期、淋巴结转移和远处转移有关(P<0.05),与年龄和Gleasonscore无关(P>0.05);GSEA分析结果 显示Plk3与前列腺癌的增殖和转移相关(FDRq<0.05)。结论 Plk3在前列腺癌中表达增高,并促进前列腺癌的进展,其可能通过促进增殖和转移发挥作用。

Abstract: Objective To investigate the expression of Polo?like kinase 3(Plk3)in prostate cancerand its clinical significance. Methods Immunohistochemistry was performed to detect the expression of Plk3 in the tissues of 68 cases of prostate cancer and 10 cases of prostate. The relationship between the expression of Plk3 and clinicopathological features was analyzed. GSEA analysis was conducted to explore relationship between gene sets of proliferation or metastasis and Plk3 expression in 498 cases of prostate cancer in TCGA database. Results The positive rate of Plk3 in prostate cancer tissues was significantly higher than that in normal prostate tissues(P=0.034). The expression of Plk3 was correlated with the pathological grade, clinical stage, tumor stage, lymph node metastasis and distant metastasis(P<0.05)but not age and Gleason score(P>0.05). The results of GSEA analysis showed that Plk3 was associated with the proliferation and metastasis of prostate cancer. Conclusion Plk3 is up?regulated in prostate cancer and promotes the progression of prostate cancer, which may play a role in promoting proliferation and metastasis.
