STAT3信号通路对大肠癌细胞VEGF表达的影响

摘要: 目的 观察STAT3信号通路对大肠癌HT-29细胞生长及VEGF表达的影响。方法 pRNAT-shSTAT3重组质粒瞬时转染大肠癌HT-29细胞,实时定量PCR、Westernblot检测STAT3的表达情况,MTT检测细胞的生长情况,实时定量PCR、ELISA检测VEGF的表达。结果 pRNAT- shSTAT3重组质粒转染大肠癌细胞后,STAT3表达明显降低,细胞生长能力明显减弱,VEGF表达明显降低,与对照组相比差异显著(р<0.01)。结论 干扰STAT3基因的表达可以有效抑制大肠癌HT-29细胞的生长及VEGF的表达。

Abstract: 【Abstract】 Objective To investigate the effects of STAT3 signaling pathway on cells proliferation and VEGF expressing in human colorectal cancer HT-29 cells. Methods HT-29 cells were transfected with the pRNAT-shSTAT3 recombinant plasmids. Real time quantitative PCR and Western-blot were used to quantify the expression of STAT3 gene. The cell growth was assessed by MTT assay. VEGF expression was detected by Real time quantitative PCR and ELISA. Results STAT3 shRNA could inhibit the expression of STAT3 gene in HT-29 cells. In the transfected group with the recombinant plasmid, the proliferation of HT-29 cells was suppressed and VEGF expression was inhibited, which was significantly different from that of the control group(р<0.01).Conclusions STAT3 singling pathway in human colorectal cancer cells can inhibit cells proliferation and VEGF expression greatly.
